The fractionation of cholesterol esters by silicic acid chromatography.

چکیده
Within recent years, the technique of silicic acid chromatography to separate complex mixtures of lipides has been developed widely (1-3). At the present time it is possible to resolve some eight different classes of lipides ranging from hydrocarbons to phospholipides on a single column (4). In addition, this procedure has been used to separate classes of phospholipides from one another (5). Our particular interest in the cholesterol ester fraction (6, 7) has led us to investigate the resolution of individual cholesterol esters by this means, and a method for obtaining four major categories has been developed. The technique compares favorably with destructive methods of analysis (i.e. ester hydrolysis and analysis of fatty acid components) while preserving the intact ester. Its application to model compounds and biological mixtures is illustrated.

The comparison of lipids between dry and wet types of cerumen.
The lipid fraction occupying about 20 to 30 per cent of the ceruminal volume was extracted by the fractionation method previously reported. The lipid fraction from the dry and wet types of cerumen collected from the Japanese children was further separated into several kinds of lipid component by silicic acid column chromatography. Identification of the respective lipid components was performed ...
متن کاملCholesterol ester fatty acids in serum and liver of normal and lymph-fistula rats.
Data on the normal distribution of fatty acids in the cholesterol ester fraction of rat plasma and liver and on factors which may influence their distribution are extremely limited at the present time. The information available has been obtained by alkali isomerization and silicic acid and paper chromatography techniques. These methods do not provide an adequate identification and estimation of...
متن کاملDirect determination of the linoleate/oleate ratio in serum cholesterol esters by liquid chromatography.
We describe a convenient method for the direct determination of the serum cholesterol linoleate/cholesterol oleate (L/O) ratio by reversed-phase "high-performance" liquid chromatography. After removal of phospholipids by silicic acid chromatography, a serum extract is analyzed on a 5-micrometers particle size Ultrasphere-ODS column, eluted isocratically with acetonitrile/isopropanol (30/70, by ...
متن کاملSeparation of tissue cholesterol esters and triglycerides by silicic acid chromatography.
bUnequivocal class separations of tissue lipids are important particularly whcn high temperature gas Chromatography is used at the same time for the determination of the long-chain acid composition of major lipid classes. This problem was studied most recently by Hirsch and Ahrens (1) ; their procedurc involves silicic acid column chromatography with clution by ether-petroleum ether mixtures fo...
متن کاملFatty Acid Components of Rat-tissue Lipids.
1. The lipids of rat heart, kidney, skeletal muscle and liver were separated by chromatography on silicic acid into cholesterol ester, triglyceride, free-fatty acid and phospholipid fractions. 2. The fatty acid compositions of these fractions were determined by gas-liquid chromatography. 3. Palmitic acid was always present in highest concentration in the cholesterol ester fraction; oleic acid w...
